How to make "256 color PNG with transparency" by PIXIA
Let's take this image to the left as a sample.
"256 color PNG with transparency" needs to choose one color in the image as the transparency color. But, in this sample image, the color of the part I want to make transparent and the color of the part I do not want to make transparent are the same: white. If I just simply make "white" of the background, the body gets transparent, too.
So paint the part you want to make transparent with a color which is not used in the image so that making that part transparent will not lead to making other parts transparent.
(Keep the number of colors used in the image less than 256 at this stage.)
Here is the configuration dialog.

Switch to "256 Palette Color" since you need this option to make 256 color PNG with transparency. Check "Transparent" option, and choose the place of the color you want to make transparent: one of the four corners of the image can be selected from the pulldown. You can check "Interlace" if you want to make interlace PNG.
Here is the result. Transparency is neatly set, isn't it?

How to make "Full color PNG with transparency" by PIXIA
Full color PNG with transparency can be achieved with the original feature of Pixia. Use the transparency channel to make the background part of the image transparent.
The easiest way of making it transparent is to use the "Eraser". Checked part of the image is considered to be transparent.
Here is the configuration dialog.

Switch to "True Color" and check "Transparent" box. Set "Filter" setting as you like, but here is a tip: Choose "ALL_FILTER" for images with simple colors like paintings, and choose "FILTER_NONE" for images with a lot of details such as Photographs. I recommend you try both and choose the better result. You can check "Interlace" if you want to make interlace PNG.
Here is the result. You cannot see the result of transparency here unless you use a browser that supports full color PNG with transparency.
